Three-Dimensional Platform-Dependent Modulation of Stemness and EMT-Associated Gene Expression in Primary and Metastatic Uveal Melanoma Models. (PubMed, Exp Eye Res)
Three-dimensional culture architecture was associated with differential modulation of stemness- and epithelial-to-mesenchymal transition-associated transcription in uveal melanoma. Platform selection and tumour origin were associated with differences in the phenotype observed in vitro and should be carefully aligned with specific biological endpoints when modelling disease progression.

Single-cell transcriptomic analyses further resolved cell-type-specific expression patterns, distinguishing tumor-intrinsic from immune-restricted expression profiles. Collectively, our findings establish CD70, CD80, and TIGIT as integrative biomarkers of tumor progression, immune contexture, and therapeutic response, providing a rationale for their clinical exploitation in precision immuno-oncology.

Pegargiminase Suppresses the Fanconi Anemia Pathway and Promotes Melphalan-Induced DNA Double-Strand Breaks in Uveal Melanoma. (PubMed, Pigment Cell Melanoma Res)
ADI-PEG20 downregulated multiple genes of the Fanconi anemia pathway and synergized with melphalan to increase DNA double-strand breaks. Melphalan and pegargiminase is a rational new drug combination that warrants clinical testing in uveal melanoma.

A Macrophage/Monocyte-Related Four-Gene Signature for Prognostic Assessment of Uveal Melanoma: BTBD6, C2CD4B, CCL24, and S100A4. (PubMed, Hum Mutat)
A significant negative correlation between RiskScore and the IC50 of XMD8-85, lapatinib, roscovitine, salubrinal, bexarotene, LFM-A13, FTI-277, and TGX221 chemotherapeutic agents was further noticed. In this study, we computationally identified genes associated with both disease progression and macrophage/monocyte-related characteristics in UVM and constructed a prognostic risk model with predicted immune infiltration patterns. These findings generate testable hypotheses that may inform future experimental studies on the immune mechanisms underlying UVM.

Ipilimumab combined with nivolumab has demonstrated superior outcomes, earning a median overall survival surpassing 70 months...The advent of new ICIs, such as relatlimab, introduces additional complexities in decision-making for first-line therapy...The FDA approval of tebentafusp and lifileucel has demonstrated potential in extending the survival time of patients with metastatic uveal melanoma...It aims to facilitate informed treatment decisions and enhance understanding of the multifaceted challenges in effectively managing advanced melanoma. As new therapies emerge, continuous education and adaptability will be key to optimising patient outcomes.

Metastasis-related genomic aberrations and evolutionary trajectory in uveal melanoma. (PubMed, Exp Mol Med)
Notably, early 6p gain was frequently exclusive of BAP1 aberrations and associated with a favourable prognosis, suggesting that metastatic potential in UM may be established early during tumorigenesis. Collectively, our findings provide molecular changes contributing to tumorigenesis and metastatic progression in UM and offer potential biomarkers for early detection and prognosis prediction.

Choroidal melanoma edge recurrence 13 years after I125 plaque brachytherapy. (PubMed, Am J Ophthalmol Case Rep)
This case illustrates a rare late local recurrence of choroidal melanoma with different appearance from the original tumor. It also highlights the diagnostic and prognostic role of transretinal biopsy in confirming the nature of the lesion as a recurrence and the option of proton beam radiation as salvage therapy.
