Co-clinical CT radiomics pipeline to establish candidate imaging biomarkers for colorectal cancer. (PubMed, Eur J Radiol)
Orthotopic KRAS-mutant xenograft models (LOVO-Luc2 (N = 52) and SW480-Luc2 (N = 52)) were treated with standard-of-care regimens (FOLFOX, bevacizumab, or combination) and longitudinally imaged by CT (N = 104 tumour scans, N = 156 liver scans collected over 4 timepoints)...The most predictive features, GLSZM Gray Level Non-Uniformity, GLRLM Run Length Non-Uniformity Normalized, and GLDM Small Dependence Emphasis, were significantly associated with metastatic burden and survival in a clinical CRC cohort (N = 41), indicating species conservation and translational relevance. Collectively, these data demonstrate that preclinical CT radiomics can identify quantitative imaging features associated with treatment sensitivity and early metastatic progression, supporting translational potential.

GALNT5 drives colorectal cancer progression and chemoresistance via PI3K/Akt/ABCC1 axis. (PubMed, J Cancer Res Clin Oncol)
GALNT5 facilitates malignant progression in CRC by promoting O-GalNAc glycosylation of EGFR and thereby activating the PI3K/Akt pathway, and confers resistance to L-OHP and 5FU through upregulation of ABCC1. These findings suggest GALNT5 as a potential diagnostic biomarker and a promising therapeutic target for overcoming chemoresistance in CRC.

Early Metastatic Relapse in Resected Stage IB KRAS G12D Pancreatic Ductal Adenocarcinoma: Limitations of Anatomical Staging. (PubMed, Cureus)
Despite apparently favorable pathological staging and adjuvant FOLFIRINOX chemotherapy, the patient developed early biochemical progression with rapidly rising carbohydrate antigen 19-9 (CA 19-9) levels, followed by widespread metastatic dissemination involving the liver, lung, spine, skeletal muscle, and multiple visceral sites...This case highlights the limitations of anatomical staging in PDAC and emphasizes the prognostic importance of tumor biology, including KRAS mutation status, lymphovascular invasion, and perineural invasion. It also demonstrates the potential limitations of CA 19-9 as a solitary marker of treatment response in biologically aggressive disease.

Prognostic markers, quality of life (QoL) and value of health (V-He) in advanced biliary cancers (ABC) treated with second-line active symptom control (ASC) alone or ASC with oxaliplatin-5-FU chemotherapy (ASC + FOLFOX) in the randomised phase III, multicentre, open-label ABC-06 clinical trial. In the setting of second-line FOLFOX for ABC, elevated tumour markers impacted on prognosis and could be utilised as stratification factors in future clinical trials. The addition of FOLFOX did not imply QoL deterioration, and did, in fact, preserve some of the status in several domains.

Metastatic appendiceal mucinous adenocarcinoma presenting as bilateral ovarian masses mimicking advanced ovarian cancer: a case report. (PubMed, Ewha Med J)
She was started on fluorouracil, leucovorin, and oxaliplatin chemotherapy. Metastatic appendiceal carcinoma can closely resemble primary ovarian malignancy. Bilateral mucinous ovarian tumors with elevated CEA levels and poor chemotherapy response should prompt evaluation for a gastrointestinal primary tumor.

Tumor regression grade for prognosis of patients with colorectal cancer liver metastasis undergoing conversion therapy (PubMed, Zhonghua Wei Chang Wai Ke Za Zhi)
The conversion therapy regimen was mainly FOLFOX, FOLFIRI, or combined with targeted therapy...A decline in serum carcinoembryonic antigen (CEA) level by more than 50% (HR=0.68, 95%CI: 0.47-0.97, P=0.027)was an independent protective factor, whereas the therapeutic response evaluated by the RECIST criteria was not an independent influencing factor. Postoperative pathological TRG is more accurate than the preoperative RECIST criteria in predicting the prognosis of patients with CRLM after conversion therapy.

A clinical study of the CALLY index and HALP score for evaluating response to FOLFOX in colorectal cancer with lung metastases. (PubMed, Am J Transl Res)
CALLY and HALP scores are effective predictors of chemotherapy response and survival in CRC patients with pulmonary metastases receiving FOLFOX. Their combined application enhances risk stratification and may assist in individualized treatment decision-making.

Real-World Survival and Toxicity Outcomes of Fluorouracil With vs. Without Leucovorin in Metastatic Colorectal Cancer. (PubMed, J Gastrointest Cancer)
In this large real-world cohort, leucovorin significantly improved survival in patients with metastatic colorectal cancer receiving fluorouracil but was associated with increased treatment-related toxicity. These findings support the continued use of leucovorin as a key component of fluorouracil-based chemotherapy regimens.

Balloon-occluded hepatic arterial infusion for unresectable hepatocellular carcinoma: a phase II trial interim analysis. (PubMed, Front Oncol)
