High-risk patients showed increased sensitivity to chemotherapeutic agents including Gallibiscoquinazole, Cisplatin, Axitinib, and Zoledronate. The anoikis-based risk model may serve as a useful prognostic tool for ACC. ARGs may contribute to ACC progression and are associated with immune microenvironment features, providing a potential basis for further mechanistic and translational studies.

The Effects of Bisphosphonates Used in Osteoporosis Treatment on Breast Cancer: Analysis with Integrative Bioinformatics Methods, DFT, ADMET and Molecular Docking Analysis. (PubMed, Biology (Basel))
This study evaluated the structural, electronic, pharmacokinetic, and receptor-binding properties of three bisphosphonate derivatives, alendronate, risedronate, and zoledronate, to investigate their therapeutic relevance in osteoporosis and breast cancer. Network analyses highlighted pathways related to mevalonate metabolism, hormone signaling, angiogenesis, extracellular matrix remodeling, and the RANK/RANKL/OPG axis. These findings support the potential repurposing of bisphosphonates, particularly zoledronate, for breast cancer-associated bone disease.

Bisphosphonate zoledronic acid blocks secretory autophagy and inhibits bone resorptive functions in osteoclasts. (PubMed, Autophagy)
In vivo studies showed that osteoclast-specific rab7 conditional knockout mice exhibited osteopetrotic phenotypes. These findings provide insights into how BPs disrupt osteoclast function by blocking autophagy and suggest that targeting autophagy in osteoclasts could be a potential therapeutic approach for bone-related diseases.

Zoledronic acid was added for osseous disease...This case highlights an unusual presentation of adult LCH with distal lower-extremity and tarsal bone involvement, as well as hypothalamic-pituitary disease preceding osseous diagnosis by two years. It underscores the importance of considering LCH in adults with unexplained multifocal lytic bone lesions and endocrine dysfunction, and it demonstrates that durable disease control can be achieved with systemic therapy.

TNFAIP3 in M2 Macrophage Attenuates Subretinal Fibrosis in Laser-Induced Murine Model. (PubMed, Invest Ophthalmol Vis Sci)
Peripheral blood monocyte-derived macrophages in mice were depleted through intraperitoneal administration of clodronate liposomes...Our findings demonstrated that TNFAIP3 in macrophages modulates subretinal fibrosis of CNV lesions by regulating M2 polarization through the NF-κB/Spi1/C/EBPβ signaling pathway. These results suggest that targeting TNFAIP3 in M2 macrophages might serve as a promising therapeutic strategy for CNV and subretinal fibrosis.
